Some of the iSuppliers are not receiving the PO Approval notifications from the Workflow process.
Please check the workflow log for more information.
You stated some users of iSuppliers does not recieve mails, any specific user setup which differentiates these users from receiving the PO approval notification. Was this functionality working fine for these user before?
Go to System Administrator --> Status monitor --> Item type as POAPPRV and User key as PO Number and see if it is stuck somewhere...

I came to know that none of the PO Approval notifications are going to supplier. Prior to upgrade to R12 all the suppliers where getting the notifications and after the upgrade this problem has occurred.
