Line Level Location?

Looking to see if it's possible to set a line level location on a transaction using a workflow.  I need them to be different from the main location, and I need them to be set automatically.  Is this possible?

I should also mention that I only want it set for certain users or for a certain transaction form, not for EVERY transaction.
