To answer your question, we need to understand the fundamental difference between delegation and transfer.
Delegation - User A takes decision on Behalf of User B
Transfer - User A now owns the task/ decision which was initially to be taken by User B
Fundamentally, transfer means the new users authority and approval would be considered over the notification.
Delegation would mean that the authority of the actual user (A) would apply, just that someone else is clicking the button for him.

Dear Abhi, Thanks for your updates, In my case we have used the Delegation that means all POs which are transfer to Manager is routed to Director. When C=Officer send his po for Approval its available on Director screen, Director approved it but PO status is not changed to Approved, for workaround I.T dept is Forward the PO to Director. Director Approved it again then PO status is updated to Approved.
So our concern is that, should we select Transfer in vacation rule so all PO approved one time as they approved by Director.
In case of delegation, ensure the Director is a part of the Position Hierarchy same as the actual approver. This is a classic case when the Position is not in Hierarchy and an approval attempt is made.
