Calculate shipping costs via script

Due to some unique business rules, we need to run a script to assign our shipping methods after entry. For shipment methods with a realtime rate, we want to update the amount during the script rather than have a user have to edit and press the shipping calculator button. I see a lot of questions around this but not for 2.0 scripts and wonder if this is possible now
