Security roles appear to be "glitching"

Content (please ensure you mask any confidential information):

One client is in UAT and users with Journal roles cannot submit journals. If roles are removed, save setup, re-added (exactly as before), save setup, user now can submit journals. Poweruser not able to run a report (that other power users identical setup can run). Change to admin, can run report, change back to power users, now can run report. Same with data exchange. User with run integration role, doesn't have abiltity to run a data load rule. change access, save, reset access exactly as before, user can run data load rule.
