Shrinking tables and indexes!!

The Enterprise Manager recommends to shrink the index and tables.  Will it lock the tables/indexes if I click the "Shrink" button in the Enterprise Manager?  As my aplplication is a 7x24 real-time system.  Should I do it at night time and shut-down the DB services first?

For the shrink or index/tables, the datafile already occupied will not be shrinked.  The spaces free after shrinked still keep in the datafile, for the tables extended in future.  Am I right?

My database is 10.2.0.3.
