Want to overwrite the workflow mail From in the notificaiton for PO approval

We have 2 workflow mailer( say mailer1, mailer2). We would like to get the From email from different mailer id based on Operating unit in PO Approval.

Do we have any setup or I have to do workflow cutomization? We have tried setting the attribute value of #FROM_ROLE, #WFM_FROM and #WFM_REPLYTO. But it is not overwriting the value. It  is always coming from one mailer id.
