What is the process for tuning a sql statement which takes long time to run in Oracle 10g EE manual

Hi experts,

What is the procedures to tune a SQL statement which runs a long time manually and automatically? My OS is Windows Server 2008 SE and My RDBMS is Oracle 10.2.0.4 EE with tuning pack but I have no EM 12C set up ready yet.
