Changes on the source while setting up the streams.

Hi,
I have 3 streams related questions. We are using Oracle 11.2 on Linux with one directional local capture streams.
Our plan is to use 4 machines (M1, M2, M3 and M4). Two machines are local (M1 and M2 in New York for example) and
two remote (M3 and M4 in California for example). The will be configured such that M1->M2->M3->M4.
- Does setting up the streams between M1 and M2 requires that our application be down on M1 while the streams is being setup?
Our application make changes to the DB on M1.
- What about when setting up the streams between M2 and M3? Since the DB is changing on M1, it will be changing
