No Response from Application Web Server

Trying to troubleshoot this APEX version 4.0.2. At the moment all the componets are working but cannot access the apex application. From IE we always get "No Response from Application Web Server"

 

"There was no response from the application web server for the page you requested.
Please notify the site's webmaster and try your request again later. "

 

The backend server hosts two databases with apex configuration, however each setup is configured in a different way, so cannot really compare both configurations and the logs are not showing any errors that will assist in resolving this issue.

Any pointers on what to check or how to resolve this issue would be great.
