Tweaking of DB settings after it is initialized

Our production DBs, which hold a PeopleSoft CS instance and an E-Business Suite instance respectively, are in need of stability and performance boosts, after we have experienced several breakdowns and/or slowdowns in both DBs during high-demand times. Is it possible to tweak the DB settings found in the initialization files for the respective applications after the DB is initialized? Re-initializing the DB is not an option for us, since our backups are not regularly done. What options (command-line or GUI) can we use to tweak the parameters and settings for each DB, particularly in the processes, open cursors and data
