'Ask for' v 'Manual' vis-a-vis Automatic Flag on the Promotion Code

Could one of the Advanced Pricing experts clarify the difference between 'ask for' and 'Manual' Promotion code (i.e. the Automatic flag is unchecked)?  Also, if the Automatic flag is checked and the 'Ask For' is also checked is it still required to manually add the promotion code?
