can we use workday definition using published schedules

For one of the project implementation we are using published schedules. There is a requirement where daily overtime need to be calculated for night shift employees. can we use workday definition with multiple shifts so that overtime is paid to night shift employee. I am aware we can achieve it using work schedule assignment. Need to know if we can use workday definition in published schedule.
