Using AME Clarification

Please If i will use AME do i need to define new oracle users in HR and assign a supervisor to each employee before managing them in AME ?

Also if i will define new 60 users on the system in all departments so every department will create its own PRs how can i restrict these departments to see only their related PRs not other departments PRs ?

Note: we are using R12.2.7
