Messages to Supervisors

There are several messages that can be sent to supervisors within the system. I'm wondering when it says "Supervisors" is this connected to Supervisor Accounts and the users mapped to those accounts, OR is it connected to fields like "manager name" in the user's individual data?
So for example - in the following scenario would the supervisor receive a message:
- Joe Dynamite is a supervisor with 2 direct reports: Harry & Hermione
- Joe Dynamite does NOT have a supervisor account or direct report mappings through a supervisor account
- Harry & Hermione's user information each have a field for "manager name" that includes Joe Dynamite's name, and a custom field for his email address
