How to calculate value based on DFF value selection

Content (required):

User selects DFF 1 (start Date) and DFF2 (End Date). System should calculate DFF3 (No of days) value automatically based on selection of DFF 1 & DFF2 while create requisition.

We can suggest user to input DFF3 manually but there might be mistakes. So business is asking us to automate this process.

Do I need to write any query in DFF3 level. Please suggest the query.
