Can custom configuration changes made in sandbox get erased during a Production environment refresh?

Summary:

We deployed a couple of UI changes on the PO page to make 4 fields (Projects and Project Details fields) mandatory. However, we were just notified that someone internally was able to submit a PO without filling out these fields.

We were wondering if during a quarterly refresh, this UI change was overwritten, or lost. Can you confirm if custom configuration change can get erased during a Production environment refresh. If so, how do we protect the customization from not being erased.
