Update an existing SO.

I have existing Sales Orders in the system that I need to update a custom field on. A vendor sends us a file with the Sales Order number and the value to place in the custom field.

I'm looking for a little direction (and sample code is awesome) for how to load an existing SO based on order number, update the custom field and then save it.
