What do I need to know?

I want to create a Advanced Customized WebStore. Do I need to know more than HTML and Dreamweaver? I also use Dynamic Drive a lot and understand how to fit javascript into my html, but do not fully understand javascript.
Basically, what all do I NEED to know?
