Update Line Item Custom Field

We have existing sales orders. Each line item of each sales orders has a custom string field. We need to update the values of the custom field for each line.

Is there a way to specify a sales order line item to update it or do we need to access the entire sales order itemsList, update that object and then resubmit the entire itemsList?
