Unsubscribe - how do I set this?

We are moving to using NS for our email blasts.  We need to import the unsubscribes from our old system - how do we do this? 

If I need to manually unsubscribe someone from all mailings (I.e. they never want to be emailed from us) - where do I do that in NS?
