Calculate shipping on item fulfillment through script

Hello All,

I need to mimic the functionality of clicking on calculate button on item fulfillment for shipping cost calculation (rates are fetched from FedEx realtime). Item fulfillment record will be created via script, need to ensure that correct shipping cost is calculated. Can anyone suggest any ideas or way to achieve this through script.
