Marking a Location as Inventory Not Available

I currently have 5 separate locations which works out fine until I look at available inventory.

Certain locations (for instance I have one called Amazon Holding) should not be included when I am running a report to see my available inventory.

If I uncheck the Make Inventory Available for these locations can I still produce an invoice to deplete the inventory in this location. I don't want this location to be used for all sales orders only the ones created for Amazon Use.

Can this be done.
