How can I tell NetSuite to use correct tax in special circumstances?

I have a situation for which I came up with a high-level solution, but there are some details that I was hoping you could help with please.
Scenario:
We will sell a certain quantity of an item, such as 50 pens, and the SO will have only one line item. These 50 items can be shipped to different locations and scripting will take care of it. And depending on the ship-to location, some items will be taxable.
Example: Assume we sell 50 pens, and 49 are shipped to New York, and 1 is shipped to California. California pen is taxable. Imagine each pen is $1. CA pen is 50% taxable.
